NoPass devices are only needed if your main flash cart is a slot-2 device, since you need to trick it into booting the slot-2 device in DS mode. Your Acekard can boot on its own, and you can even use it as a NoPass if you ever get a slot-2 device and want to boot it in DS mode for any reason.

I have DS Lite and based what I read on its official website, R4 is the best card to get, since I can just drag and drop games into the card and run right away. Does that mean the card comes with .nds launcher?

The R4 is the worst cart to get nowadays. ALL modern slot-1 devices are drag-and-drop, and nearly all have more features and better support than the R4. Look into the Acekard 2i or CycloDS Evolution or EDGE or Supercard DS instead. Also consider if you want the cart to be DSi-compatible, which only certain devices support.

I would also like to see the answer to this question. But am I correct to presume that this can be accomplished with one of the newer slot 1 cards alone such as the M3i Zero?
